Lidocaine prevents breast cancer growth by targeting neuronatin to inhibit nerve fibers formation

چکیده
Lidocaine has been shown to inhibit the invasion and metastasis of breast cancer, but mechanism still remains unclear. This study explored relationship between lidocaine circulating seeding cancer cells from perspective nerve fiber formation. The cell lines MDA-MB-231 4T1 were subcutaneously inoculated in mice simulate tumor self-seeding by cells. was used treat these growth observed. Silver staining performed observe distribution fibers tumor-bearing tissues, immunohistochemical analysis expression levels nerve-related proteins. results showed that treatment effectively inhibited formation, down-regulated protein gene product 9.5, neurofilament, factor (NGF), neuronatin (Nnat). Overexpression NGF Nnat both could reverse therapeutic effects lidocaine. These suggest effect on inhibiting may be achieved targeting Nnat, regulating production NGFs cells, subsequently formation fibers.

Targeting G Signaling to Inhibit Prostate Tumor Formation and Growth*
Prostate cancer starts as androgen-dependent malignancy and responds initially to androgen ablative therapy. Beneficial effects of androgen ablation, however, are often temporary and the cancer reappears as androgen-independent tumor, suggesting the existence of additional factors responsible for progression of the disease. Attention has focused on receptor tyrosine kinases as the growth mediat...
متن کاملNerve Fibers in Breast Cancer Tissues Indicate Aggressive Tumor Progression
Emerging evidence has indicated nerve fibers as a marker in the progression of various types of cancers, such as pancreatic cancer and prostate cancer. However, whether nerve fibers are associated with breast cancer progression remains unclear. In this study, we evaluated the presence of nerve fibers in 352 breast cancer specimens and 83 benign breast tissue specimens including 43 cases of cyst...
متن کاملTherapeutic targeting of PELP1 prevents ovarian cancer growth and metastasis.
PURPOSE Ovarian cancer remains a major threat to women's health, partly due to difficulty in early diagnosis and development of metastases. A critical need exists to identify novel targets that curb the progression and metastasis of ovarian cancer. In this study, we examined whether the nuclear receptor coregulator PELP1 (proline-, glutamic acid-, leucine-rich protein-1) contributes to progress...
متن کاملMiR-490-5p Functions as an OncomiR in Breast Cancer by Targeting NFATc4
Breast cancer is a serious health problem worldwide in women. MicroRNAs are small non-coding RNAs of 18–25 nucleotides in length that post-transcriptionally modulate gene expression. MiR-490 has been reported as a tumor suppressor and oncomiR microRNA in breast cancer with two separate targets, NFAT and Rho. NFAT is one of the targets for miR-490 but the relationship between hsa</e...
متن کاملHuman Wharton’s jelly mesenchymal stem cells-derived secretome could inhibit breast cancer growth in vitro and in vivo
Objective(s): Controversial results have been reported regarding the anti-tumor properties of extracellular vesicles derived from mesenchymal stem cells (MSCs). The present study was conducted to evaluate whether secretome derived from Human Wharton’s jelly mesenchymal stem cells (hWJMSCs) may stimulate or inhibit breast cancer growth in vitro and in vivo.<st...
